Design Notes
During the detailed design of this part. Firstly we made an amino acid substitution K229L which has been shown in the literature to lead to higher activity through inhibiting negative regulation and therefore higher rates of aromatic amino acid biosynthesis. Secondly codon optimisation was carried out using Benchling's codon optimisation tool. Optimisation settings were set to: any GC content; avoid hairpins (20;200); and for Saccharomyces cerevisiae. Furthermore in the same tool several cut sites were forbidden, these restriction sites were BsaI (BASIC Assembly) and EcoRI, XbaI, SpeI, PstI, and NotI (BioBrick assembly).
